【背景】传统制作奶豆腐和酸性奶油(乌日莫)是内蒙古农牧地区最喜爱的食品,蕴含着十分丰富的乳酸菌资源,亟待开发利用。【目的】通过解析内蒙古农牧地区传统自制奶豆腐和乌日莫样品中乳酸菌多样性及分布特征,为优良菌株选育与利用提供资源和理论基础。【方法】采用稀释涂布法分离纯化乳酸菌,测定菌株16S rRNA基因序列鉴定种属关系,阐明乳酸菌系统发育、遗传分化及菌群结构。【结果】传统自制样品中共分离得到乳酸菌81株,主要归属于乳酸片球菌(Pediococcus acidilactici)、戊糖片球菌(Pediococcus pentosaceus)、短乳杆菌(Lactobacillus brevis)、瑞士乳杆菌(Lactobacillus helveticus)、副干酪乳杆菌(Lactobacillus paracasei)、食二酸乳杆菌(Lactobacillus diolivorans)、奥塔基乳杆菌(Lactobacillus otakiensis)、植物乳杆菌(Lactobacillus plantarum)、开菲尔乳杆菌(Lactobacillus kefir)、乳酸乳球菌(Lactoc...  相似文献

本研究旨在建立一种简便、快捷、可直观检测小反刍兽疫病毒(peste des petits ruminants virus,PPRV)抗体的检测方法。将pET-32a-N重组质粒转化至大肠杆菌(Escherichia coli) Rosetta(DE3)感受态细胞中进行诱导表达,以纯化的PPRVN蛋白免疫8周龄BALB/c小鼠,取其脾细胞与SP2/0骨髓瘤细胞进行融合,间接酶联免疫吸附试验(enzyme-linked immunosorbent assays, ELISA)筛选及亚克隆,获得了抗PPRV N蛋白的单克隆抗体。将PPRV N蛋白分别作为金标抗原及检测线(T线)包被抗原、单克隆抗体作为质控线(C线)包被抗体,组装成检测PPRVN蛋白抗体的胶体金免疫层析试纸条。结果显示:成功获得1株能稳定分泌抗N蛋白抗体的杂交瘤细胞株,命名为1F1;间接ELISA检测1F1腹水效价为1:128000;亚类鉴定结果为IgG1,轻链为kappa链。Westernblotting结果显示,1F1能与PPRV N蛋白特异性结合;间接免疫荧光(indirect immunofluorescent ass...  相似文献

The heartwood of Syringa oblata Lindl. (SO) is one of Mongolian folk medicines to treat insomnia and pain, while its pharmacological evaluation and underlying mechanism remain unclear. In this study, the sedative effect of ethanol extract of SO (ESO) was evaluated with the locomotor activity test and the threshold dose of pentobarbital sodium-induced sleep test in mice, and the hot plate test, acetic acid-induced writhing test, and formalin test in mice were used to evaluate its analgesic effect. The underlying mechanism of ESO analgesia was explored by RT-PCR and western blot analysis, which is associated with the regulation of the NF-κB signaling pathway. Besides, the main constituents of ESO were characterized by LC/MS data analysis and comparison with isolated pure compounds. The current findings brought evidence for clinical application and further pharmacological and phytochemical studies on SO.  相似文献

Cytochrome P450c17 (CYP17, 17α-hydroxylase/17, 20-lyase) plays a critical role in the production of androgens and estrogens in vertebrates. We isolated the full length cDNAs of P450c17-I and P450c17-II from Sebastes schlegeli. The cDNA sequences of P450c17-I and P450c17-II encoded 515 and 533 amino acid residues respectively. The putative P450c17-I and P450c17-II enzymes of Korean rockfish share high sequence identity with that of Japanese flounder (92% and 81%) respectively. Our current study describes that P450c17s of Korean rockfish are mainly expressed in gonads, head kidney and kidney by RT-PCR. Quantitative real-time PCR showed that the expression patterns of Korean rockfish P450c17s were developmental stage-dependency. In addition, the testosterone (T) and gonadosomatic index (GSI) levels further support the important role of P450c17-I during shift in steroidogenesis. Taken together, this study provides information about the Korean rockfish P450c17s characterization and mRNA expression as such helps in further understanding of its function in gonadal development.  相似文献

Antioxidant cellular mechanisms are essential for cell redox homeostasis during animal development and in adult life. Previous in situ hybridization analyses of antioxidant enzymes in zebrafish have indicated that they are ubiquitously expressed. However, spatial information about the protein distribution of these enzymes is not available. Zebrafish embryos are particularly suitable for this type of analysis due to their small size, transparency and fast development. The main objective of the present work was to analyze the spatial and temporal gene expression pattern of the two reported zebrafish glutathione peroxidase 4 (GPx4) genes during the first day of zebrafish embryo development. We found that the gpx4b gene shows maternal and zygotic gene expression in the embryo proper compared to gpx4a that showed zygotic gene expression in the periderm covering the yolk cell only. Following, we performed a GPx4 protein immunolocalization analysis during the first 24-h of development. The detection of this protein suggests that the antibody recognizes GPx4b in the embryo proper during the first 24 h of development and GPx4a at the periderm covering the yolk cell after 14-somite stage. Throughout early cleavages, GPx4 was located in blastomeres and was less abundant at the cleavage furrow. Later, from the 128-cell to 512-cell stages, GPx4 remained in the cytoplasm but gradually increased in the nuclei, beginning in marginal blastomeres and extending the nuclear localization to all blastomeres. During epiboly progression, GPx4b was found in blastoderm cells and was excluded from the yolk cell. After 24 h of development, GPx4b was present in the myotomes particularly in the slow muscle fibers, and was excluded from the myosepta. These results highlight the dynamics of the GPx4 localization pattern and suggest its potential participation in fundamental developmental processes.  相似文献

目的:探讨蒙药乌力吉-18对大鼠下丘脑-垂体-卵巢轴相关激素及受体的影响。方法:选取40只健康雌性未孕SD大鼠,随机分为空白组、对照组、乌力吉-18高、低2个剂量组,每组10只。空白组灌胃等体积蒸馏水,对照组灌胃逍遥丸,高、低剂量组分别灌胃2.0 g·kg-1·d-1、1.0 g·kg-1·d-1乌力吉-18,连续给药31学艺术d。采用酶联免疫吸附法测定血清促性腺激素释放激素(GnRH)、促卵泡生成素(FSH)、黄体生成素(LH)、雌二醇(E2)及孕酮(PROG)的含量;免疫组化法检测下丘脑组织促性腺激素释放激素(GnRH)、垂体组织促性腺激素释放激素受体(GnRHR)的表达;以蛋白免疫印迹技术检测卵巢组织促卵泡生成素受体(FSHR)、黄体生成素受体(LHR)蛋白表达量。以实时荧光定量PCR检测卵巢组织中FSHR、LHR基因表达量。结果:与空白组比较,乌力吉-18低剂量组可明显升高血清LH含量(P<0.05),上调下丘脑组织GnRH、垂体组织GnRHR表达及卵巢组织FSHR、LHR蛋白表达(P<0.05);乌力吉-18高剂量组可显著升高血清FSH、LH、E2含量(P<0.05),上调下丘脑组织GnRH表达及卵巢组织FSHR表达量(P<0.05),并可显著升高卵巢组织中FSHR、LHR基因表达量(P<0.05);对照组可明显升高血清E2含量(P<0.05)。结论:蒙药乌力吉-18可明显升高血清FSH、LH及E2的含量,促进下丘脑组织GnRH、垂体组织GnRHR及卵巢组织中FSHR、LHR的表达,表明乌力吉-18能够对下丘脑-垂体-卵巢轴相关激素及受体表达产生影响。  相似文献

大青沟自然保护区主要森林群落优势种的叶性状   总被引:1,自引:0,他引:1  
植物功能性状是近年来生态学研究的热点。其中叶功能性状与植株生物量和植物对资源的获得、利用及利用效率的关系最为密切。大青沟森林植物群落分布于科尔沁沙地,生境条件非常特殊,在沙沟里存在着一片茂密的森林,与周围浩瀚无垠的沙坨景观形成极为鲜明的对照。从沟底到沟顶,虽然海拔高度仅相差六、七十米,但由于距离沟底水源不同,土壤条件差异大,形成了不同的森林植物群落类型。大果榆群落、蒙古栎群落和水曲柳群落分别分布在大青沟自然保护区的沟顶、沟中和沟底。为了对大青沟自然保护区,不同环境梯度下的森林植物群落叶片功能性状进行研究,以大青沟自然保护区大果榆、蒙古栎、水曲柳3种主要森林群落为研究对象,分别测定不同群落优势种的叶厚度、比叶面积、叶干物质含量、叶大小和叶干重等5项叶功能性状,研究叶功能性状之间的关系,并对不同生长型、不同群落叶功能性状进行比较。相关分析结果表明,叶厚度与比叶面积呈极显著负相关,与叶大小和叶干重呈极显著正相关;比叶面积与叶干物质含量、叶干重呈极显著负相关,与叶大小呈显著正相关;叶干物质含量与叶大小呈极显著负相关,与叶干重呈极显著正相关;叶大小与叶干重呈极显著正相关。不同生长型植物叶片性状的分析表明,草本植物的叶干物质含量比乔木和灌木低,而其比叶面积高于乔木和灌木;对不同群落叶功能性状进行比较发现,大果榆群落和蒙古栎群落乔木、灌木叶厚度与干物质含量显著高于水曲柳群落的叶厚度与干物质含量,二者比叶面积显著低于水曲柳群落。大果榆群落和蒙古栎群落从叶功能性状的角度,它们具有较高的叶干物质含量和较低的比叶面积,体现出适应干旱生境叶片的特征。水曲柳群落呈现出低叶干物质含量、高比叶面积的特征,体现出适应湿润、土壤水分较好生境的特征。不同群落通过调节自身的物种组成,形成不同的功能性状组合来适应环境。  相似文献

目的:研究阿霉素损伤心肌细胞miRNA378与网腔钙结合蛋白(calumenin)、内质网应激相关性。方法:原代培养乳鼠心肌细胞分为6组:对照组、阿霉素组、miRNA378过表达对照组、miRNA378过表达组、miRNA378沉默对照组、miRNA378沉默组,采用免疫组化法检测细胞α-SMA蛋白;慢病毒质粒转染心室肌细胞,实时荧光定量PCR技术检测各组心肌细胞miRNA378、calumenin及葡萄糖调节蛋白78(GRP78) mRNA表达。结果:与阿霉素组相比较,miRNA378过表达组心肌细胞calumenin mRNA表达增加(P<0.01),而GRP78 mRNA表达减少(P<0.01);与阿霉素组相比较,miRNA378沉默组calumenin及GRP78 mRNA表达无统计学差异。结论:阿霉素损伤乳鼠心肌细胞是通过减少calumenin蛋白表达进而引起内质网应激,该作用通过上调miRNA378得到缓解。  相似文献
